肺穿刺标本p53、PCNA蛋白表达及DNA倍体流式细胞分析

                        ABSTRACT
  Objective: To evaluate (1) the  expression of p53, PCNA protein and DNA ploidy in percutaneous biopsy specimen of lung lesions, (2) the relationship between p53, PCNA, DNA ploidy and clinical parameters. 
  Materials and Methods: CT-guidance percutaneous biopsy of lung lesions were performed in 80 patients, The expression of p53, PCNA protein, DNA ploidy were determined by Flow Cytometer (FCM).
  Results: 1. 65 patients with lung cancer and 15 patients with benign pulmonary diseases were confirmed by pathology and clinic follow-up. The sensitivity of CT-guidance percutaneous biopsy was 95.34% in lung cancer. the diagnostic accuracy of percutaneous biopsy was 93.8% in all. 2. Between lung cancer and benign cases, the differences of the expression of  p53, PCNA protein, DNA ploidy were significant(P<0.01). The expression of p53 ,PCNA protein and DNA ploidy were not related to pathological type or stage of disease. 3. The expression of p53, PCNA protein were correlated with the CT signs of deep lobulation, spine(P<0.05). DNA ploidy was correlated with deep lobulation and mediastinal lymph nodes metastasis(P<0.05), but it were not related to tumor size, the signs of spiculation, pleural indentation.
  Conclusion: Flow Cytometer play an valuable role in determining the expression of p53, PCNA protein and DNA ploidy in the percutaneous biopsy specimen of lung cancer. The expression of p53, PCNA and DNA ploidy protein may be useful in diagnosis of lung cancer, and it provide us to realize the relationship between characteristic of molecule biology and clinical parameters of lung cancer.
