Abstract

Objective:To evaluate the therapeutic effect of VEGF plasmid in cerebral infarction of dog by MR-DWI.

Methods:18 adult dogs were divided into three groups randomly after the focal cerebral ischemic infarction model was produced. Each group was divided into two subgroups,the experimental group was given VEGF plasmid 600μg and the control group was given the same amount of saline. MR scan was performed repeat-edly. After various survival times, specimens were selected and stained with HE or immunohistochemistry. The microblood vessel counts, VEGF im-munostained cells and ADCr were calculated and analyzed by SPSS 10.0.

Results:The signal intensity ofinfarcted tissue on DW images was markedly higher than that of normal brain tissue after stroke, ADC value declined to ( 5.61 ±1.39) mm 2/s,only 43 % of the contralateral. The ADCr of the two groups were significantly different at the end of two weeks ( F =0.032,Ρ= 0.009); there was significant positive correlation relationship between ADCr and microvessel counts (MVC) ( r = 0.679,Ρ <0. 01).

Conclusion:The application of VEGF plasmid in cerebral infarction of dog can improve the collateral vessel formation; MR -DWI combined with traditional MR sequences can assess the therapeutic effect of VEGF plasmid quantitatively or semiquantitatively.
